The S1-S2 FCS is both necessary for efficient transmission through the respiratory route in most animals, and disables the ability for a Sarbecovirus to form reservoirs within said animals by inactivating intestinal shedding of the virus and preventing the proper reinfection of animal hosts (or even human hosts before the roll-out of Adenovirus-based vaccines https://www.medrxiv.org/content/10.1101/2020.08.24.20179457v2). This makes such a virus a perfect candidate for self-spreading vaccines as it is both spread efficiently (for the animal models they tested in North America) and disappears from the target population once the population have been fully immunized. P681 is necessary for the efficient production of viral stocks within VERO E6 cells and PRRARS was the most lung- and airway- compatible FCS that the VERO E6 cell line will still accept and grow efficiently. Passage of the virus within airway cell line models while preparing the stock in VERO E6 turn an otherwise canonical RRRARRS sequence into PRRARS. (So will VERO passage with frequent plaque purification and western blotting that ensure the Spike protein produced is still cleaved)
